What is AKTA avant?
ÄKTA avant is a preparative chromatography system intended for method and process development (Fig 1). ÄKTA avant 150 system, with flow rates up to 150 ml/min, is designed for scaling up to larger columns. UNICORN™ software is used to control automated processes on ÄKTA avant to increase productivity and efficiency.
What is Akta chromatography?
ÄKTA pure is a flexible and intuitive chromatography system for fast purification of proteins, peptides, and nucleic acids from microgram levels to tens of grams of target product.
How do you use Akta?
Akta Pure General Protocol V. 1
- Connect buffers and prime pumps. Complete a pump wash with water on all pumps (A and B).
- Connect column. In Unicorn, go to “Manual Instructions” > “Alarms” > “Alarm pre column pressure.” Set the high alarm pressure limit that is appropriate for the column.
- Prime and fill sample loop.
What is the difference between FPLC and HPLC?
HPLC is an analytical technique used primarily for quantitative and qualitative analysis of liquid samples. On the other hand, FPLC is a purification technique which is used to separate protein mixtures and to collect the separated components of interest.
How are buffers prepared in the Akta avant system?
Buffers can be prepared in the ÄKTA avant system using the BufferPro tool, where the input is the desired pH, buffer, and salt concentration. BufferPro then controls the quaternary valve and automatically adjusts the appropriate proportions of acid, base, salt, and water. BufferPro contains 19 different buffer systems to choose from.
What is the purpose of Akta avant chromatography?
ÄKTA avant is a preparative chromatography system designed for fast and secure development of scalable methods and processes.
How to perform two step purification in Akta?
Introduction This cue card describes how to configure ÄKTA pure, set up methods, and perform a fully automated two-step purification using two one-step UNICORN™ methods in a method queue. ÄKTA avant can be configured applying the same principle. The fully automated method is suitable
How is Scouting used in the Akta avant?
Whereas pH feedback is available in the BioProcess IC System, scouting can be used in the ÄKTA avant to explore how the pH varies as a function of different relative proportions between the acid and the base. The scouting function makes it possible to do multiple runs in an automated way to build a pH titration curve (Fig 4).
